I have about 27,550 emails currently in my inbox. My iPhone is set to get 200 of the most recent emails, the maximum the iPhone will support.
Will Nuevasync be able to handle this? Can I anticipate any problems, short of my own mail server having a problem?

Hi, one thing to clear up first is that the 200 recent messages setting on the phone applies only to the phone's IMAP client. When you sync, a different limit applies inside our service. It happens to default to 200 messages at present, but we're making the limit user-configurable.
So, I like to keep a clean inbox, but I have 100k+ messages in my trash folder and just tested opening that folder on a device. The initial sync (the most costly part of the process) completed in 20 seconds or so.
As you surmise, if your mail server has trouble processing the searches we sent it, then there could be trouble.
If you sign up and subsequently have problems due to folder size, we'll either fix the problem or find some other way to make you happy.

Any ETA on this feature to the public? & Will this eliminate the need to perform server side searches on our mailboxes/folders, because we will be storing the emails locally on the phone?

The UI is done, but we need to add a way for users to force a resync from the web site (otherwise some of the settings you can change won't actually take effect). I hope to have that done tomorrow.
I don't think this is a great substitute for server search, but it may help in some cases since you can have more messages. Some users may prefer fewer messages downloaded -- the native IMAP client only fetches 50 by default.

I agree Server search is a necessity. I couldn't imagine having 25k messages on my phone. Also for me historical email search is essential.
